A method for production and utilization of fluorophore-labelled oligonucleotides to identify very localized regions of chromosomes. The method of degenerate oligonucleotide primed PCR (DOP-PCR) generates an assortment of characteristic fragments by use of a primer with arbitrary but fixed 3'- and 5'-ends, but randomized internally, so that it will find an appropriate number of hybridization sites on both strands of a source DNA to generate a convenient number of PCR fragments; in a selection step, the DOP-PCR products are annealed to carefully selected and characterized chromosome fragments fixed to a solid support, washed to remove non-hybridizing fragments, and the successfully annealed oligonucleotides eluted and labelled to serve as 'paint' that will colour and thus identify the chromosome region that selected them.Bailey, D.M.D., Carter, N.P., de Vos, D., et al. A technique for screening bacterial colonies for those that contain a desired polynucleotide sequence. A plate is exposed to a labelled oligonucleotide probe with a sequence complementary to part of the desired sequence which thus labels clones with that sequence. In a variant technique, plaque hybridization, phage vectors are screened for the desired polynucleotide sequence. Related tool: molecular biology tools

Descriptive of procedures or processes that generate a diversity of molecules (e.g. nucleic acid or protein sequences; related chemical structures) that can be screened for a desired property. The collection of molecules is termed a library. (see also small molecule library)
