An epithelial cell of a lower animal in which pigment granules can be physically moved to effect colour changes.
A technique for protein separation that uses high-pressure liquid chromatography followed by sodium dodecyl sulphate (SDS)/polyacrylamide-gel electrophoresis. The effluent from a reverse-phase column is mixed with SDS and a reducing agent and applied to a polyacrylamide slab gel; the resulting gel shows two-dimensional separation, by polarity in one dimension and by molecular mass in the other. The localization of genetic traits to regions of chromosomes, by linkage studies (e.g. the association of haemophilia with the X chromosome) and by in situ hybridization to fluorescent probes.
In positional cloning, an alternative to chromosome walking for finding a gene. Clones of genomic DNA are fragmented so as to include both the target gene and a closely linked marker and are screened to select ('land on') those clones that contain the target gene. (= coincidence painting (chromosome painting))
A strategy for mapping and sequencing a chromosome. Large restriction fragments are generated and, by Southern blotting with an RNA or cDNA probe, a single starting point is identified. New probes are synthesized from sequences of the same fragment that are adjacent to the starting point, and are then used to identify different restriction fragments adjacent to that which bears the starting point. The procedure is used repetitively, working towards the ends of the chromosome and away from the starting point. Related tool: real time pcr
