What the Data Say

• Case study #1: Successful Synthesis & Assembly of a 50 kb Gene

  Objective: To synthesize a gene (Homo sapiens) with 50,245 bp in length, located on chromosome Y.

  Strategy: (a) Divided the sequence into 10 x 5 kb fragments containing Ascl restriction sites at 5’ and 3’ ends with 200 bp overlap regions (Fig. 1); (b) all fragments were cloned in pUC57 vector and then transformed in yeast for assembly; (c) after screening (Fig. 2), selected clones were subjected to restriction enzyme digestion and NGS sequencing for final validation.

  Results: Results from the gel electrophoresis of digested gene (Fig. 3) and also NGS sequencing (Fig. 4) demonstrated that the 50 kb gene was synthesized without any error and at full length.

• Case study #2: Successful Synthesis & Assembly of a 100 kb Gene

  Objective: To synthesize a gene (combination of Bat coronavirus genome and partial Escherichia coli genome) with 100,620 bp in length.

  Strategy: (a) Divided the sequence into 4 x 25 kb fragments with each 25 kb fragment first assembled from 5 x 5 kb fragments containing 500 bp overlaps and NotI site (Fig. 1); (b) all fragments were cloned in pUC57 vector followed by transformation in yeast for assembly; (c) after screening (Fig. 2), selected clones were subjected to NGS sequencing for final validation.

  Results: Results from the gel electrophoresis of digested gene (Fig. 3) and also NGS sequencing (Fig. 4) demonstrated that the 100 kb gene was synthesized without any error and at full length.

• Case study #3: Successful Synthesis & Assembly of a 200 kb Gene

  Objective: To synthesize a gene (combination of Bat coronavirus genome, partial Vibrio natriegens genome and partial Vibrio natriegens genome) with 202,155 bp in length.

  Strategy: (a) Divided the sequence into 7 x 25 - 35 kb fragments with each fragment first assembled from several 5 kb fragments containing 500 bp overlaps and NotI sites (Fig. 1); (b) all fragments were then cloned in pCC1 or pUC57 vectors followed by transformation in yeast for assembly; (c) after screening (Fig. 2), selected clones were subjected to restriction enzyme digestion and NGS sequencing for final validation.

  Results: Results from the gel electrophoresis of digested gene (Fig. 3) and also NGS sequencing (Fig. 4) demonstrated that the 200 kb gene was synthesized without any error and at full length.
