photoaffinity labeling

Photoaffinity labeling is a technique used to attach "labels" to the active site of a large molecule, especially a protein.

Photoaffinity labeling (PAL), using a chemical probe to covalently bind its target in response to activation by light, has become a frequently used tool in drug discovery for identifying new drug targets and molecular interactions and for probing the location and structure of binding sites.

To be more specific, photoaffinity labeling is a variant of affinity labeling in which the covalent attachment of a ligand to its site is affected by the illumination of a light-sensitive chemical group on the ligand, often an azido group.

Photoaffinity labeling (PAL) is another successful and widespread strategy that facilitates the implementation of target-fishing with AfBP probes. It contains a photoactivatable functional group, such as diazirine or benzophenone.
