Polyacrylamide gel electrophoresis

Polyacrylamide gel electrophoresis (PAGE) is a method commonly used to separate proteins. Polyacrylamide, or polymerized acrylamide, forms a porous gel matrix through which proteins migrate according to their structure and overall charge under an applied electrical field. Under denaturing conditions, by adding sodium dodecyl sulfate (SDS-PAGE) and a reducing agent (i.e., 2-mercaptoethanol), proteins separate based on the length of the polypeptide chain.

Polyacrylamide gel electrophoresis can also be used to separate DNA fragments, both double and single-stranded DNA. Several advantages of PAGE for DNA separation include higher resolution, analysis of larger DNA quantities, and recovery of high purity DNA to enable downstream applications (e.g., embryo microinjections).
