Dr. Xu: Many papers have discussed this topic. Of course, it highly depends on which protein you want to express. I remember there was a very nice paper published in 2019 comparing CHO-S, CHO-K1 and CHO-DG44 for protein expression capacity. Their data demonstrated that the addition of feed concentrates substantially influenced cell accumulation by favoring one CHO host over the other regarding their specific nutrient demands. However, it seems that CHO-K1 always results in the highest titer for mAb expression, while CHO-S seems to be a better choice when expressing protein for biomass synthesis.

The Workhorse of Protein Expression in Pharmaceutical Industry – CHO Expression Platform
In this webinar, Field Application Scientist at GenScript Europe, Dr. Shuting Xu, gave a quick review of the common protein expression systems and introduced the history of CHO cell line in protein expression. And then she talked about how to accelerate the research of protein and antibody. According to the characteristics comparison of the 3 main expression systems, we may conclude that the mammalian expression system presents several advantages:

Comprehensive post-translational modification
So the mammalian expression system is the most suitable platform to produce secretory proteins and antibodies.
For the current antibody therapy strategies against cancer, the applications can be divided into 4 directions depending on the decorations of antibodies:

HEK293 and CHO expression systems are commonly used for transient expression system, so what are the advantages of CHO cell compared with HEK293 cell?

What are the disadvantages of CHO expression system?
Dr. Xu: CHO system is not perfect as I mentioned in my talk, I think when we talk about disadvantages is always compared with other systems.
Compared to the E.coli system, the CHO cell is always slightly more expensive.Compared to HEK 293 cell, on the experience when we express the same proteins, could be a slightly taking longer time.Compared to HEK 293 cell, it's a post-transcriptionally modification.So these are the disadvantages of CHO cell expression system.

There're a lot of factors co-determining the expression level. Expression vectors, cell lines, and the sequence itself. I am wondering whether GenScript can do any codon optimization for protein sequences and whether it helps.
